Preparation

Streak out yeast and grow until small colonies form.

Procedure

Place 5 ul of water into a PCR tube.

Stick a pipette tip into the colony to pick up a small number of cells. Smash the pipette tip into the cells.

Freeze the PCR tube for 10-15 min.

Microwave the PCR tube for 1 min on high power.

Freeze the PCR tube for 10-15 min.

Prepare 45 ul of the PCR mix.

Add the PCR mix to the tube, mix, spin down, and PCR as normal.
